Semax is the heptapeptide Met-Glu-His-Phe-Pro-Gly-Pro, and its construction explains what it is for. The first four residues reproduce the ACTH(4-7) fragment of adrenocorticotropic hormone. The last three, Pro-Gly-Pro, replace the Arg-Trp-Gly that the parent fragment carries.

That one substitution does two things at once. The proline-rich tail creates a conformational constraint that aminopeptidases and carboxypeptidases struggle to recognise, so the molecule survives in plasma far longer than the parent fragment. And it removes the corticotropic activity: Semax does not drive cortisol release from the adrenal cortex the way full-length ACTH does. Researchers get the central activity attributed to the ACTH fragment without the hormonal axis attached to it.

Published rodent work has measured activation of dopaminergic and serotonergic systems, expression of BDNF and NGF, and behaviour in learning and attention tasks. Ischaemia and neuroprotection models appear frequently, following the compound’s development in Russia for that research context.

Selank is the sibling compound. It uses the identical Pro-Gly-Pro strategy on a different parent peptide, which makes the two a natural pair for comparing what the extension contributes against what the parent sequence contributes.
